An acute dose of 2,000mg/kg Deer Velvet Antler to rats (human equivalent dose of 320mg/kg) has failed to show toxic signs over 14 subsequent days of observation, and a 90 day trial with daily dosing of 1,000mg/kg did not show any significant toxicological symptoms of haemotological signs; a decrease in liver weight was noted in males, but under histological examination it appeared to be benign. Another rat toxicological study using 10% of the diet as Deer Velvet Antler during gestation and after birth noted that there were no apparent teratogenic effects on the rat pups and that serum AST (indicative of liver damage) was actually decreased 50% relative to control with no effect on γ-GT (another liver enzyme).
No direct reports of chronic wasting disease (CWD) related to deer velvet supplementation have been published. However, several Web sites contain disclaimers mentioning the possibility of the disease being present in antler products. The CDC has not yet found a relationship between CWD and any neurological disease that affects humans with deer velvet use.
On January 30, 2013, Vijay Singh professional PGA Tour golfer was caught unaware and openly admitted to the personal use of deer antler spray which contained a banned substance at the time. A week later the World Anti-Doping Agency (WADA) lifted the ban on deer antler spray, but with urgency, "Deer Antler Velvet Spray may contain IGF-1 and WADA recommends therefore that athletes be extremely vigilant with this supplement because it could lead to a positive test."  The consensus opinion of leading endocrinologists concerning any purported claims and benefits "is simply that there is far too little of the substance in even the purest forms of the spray to make any difference,"  and "there is no medically valid way to deliver IGF-1 orally or in a spray."

Luteinizing hormone (LH) and follicle-stimulating hormone are hormones that stimulate the sex glands. The stimulation of the sex glands is pivotal for sexual reproduction. Without the presence of the gonadotropin hormones, the sexual organs will fail at their purpose and the individual will be considered infertile. As the deer antler velvet promotes an increase in LH, the LH causes the testes to produce more testosterone.
Russian and Japanese researchers have conducted experiments using deer antler extract and found that it appears to lower blood pressure in both human subjects and laboratory animals. A series of clinical case studies (Albov, 1969) were conducted in which the effects of Pantocrin on cardiac patients were assessed. In one test involving 32 subjects with high blood pressure caused by cardiac disease, early onset menopause or obesity, blood pressure was lowered in 81% of patients. In another study involving 13 patients with hypotension caused by disorders of heart muscle activity, blood pressure was lowered for 84% of patients.
Deer antler products contain mostly amino acids (that form proteins) along with growth factors, which are poly-peptide bonded amino acid chains. (4) The most abundant growth factor is IGF-1. Unfortunately, the potential problems with IGF would seem to negate any of these theoretical benefits. It has been shown that improper use of hormones such as dehydroepiandrosterone (DHEA), androstenedione, and human growth hormone may increase the risk for development of prostate cancer or promote the growth of existing prostate cancer by raising IGF-1 levels. Therefore, men who are taking supplements with IGF in it (or those that raise IGF levels) could theoretically be putting themselves at an increased risk for prostate cancer. Again, it hasn’t been rigorously studied so it’s impossible to know for sure, but if you have any risk factors for prostate cancer, it’s probably best to avoid taking this supplement.

In a randomized, placebo controlled test in 2004, researchers at the University of Alberta, Canada, placed 18 males from the Edmonton Police Force into a 9 week strength training program. The results showed that deer antler velvet increased the strength and endurance of the subjects relative to the control group. The researchers found that use of deer antler velvet significantly increased blood plasma testosterone levels.
