In an ovalbumin sensitized mouse model, 4 weeks of Velvet Antler at 2.5-10mg total (weight of mice not given, assuming 20g this equals 125-500mg/kg or 10-40mg/kg for humans) was able to reduce total Immunoglobulin E (IgE) and Ovalbumin-specific IgE at 14, 21, and 28 days. When challenged with methacholine and subsequently having their airway power measured, it appeared that Velvet Antler exert anti-asthmatic effects in regards to allergies.

Reports claim deer antler helped Ray Lewis overcome his recent triceps tear, and Vijay Singh has admitted to using a spray supplement. University of Alabama football players also allegedly used deer antler sprays leading up to the 2013 BCS National Title Game. Whether or not those reports are true, one thing is certain: There’s not much proof that deer antler is a performance enhancer or a miraculous healer.
All groups experienced a significant improvement in strength. But the deer antler powder group showed the greatest increase in isokinetic knee extensor strength and endurance. However, it’s possible that this was due to exercise program rather than using the deer antler powder. None of the men in any group showed evidence of endocrine, red cell mass or VO2max changes. Therefore, researchers concluded that the “findings do not support an erythropoetic or aerobic ergogenic effect of deer antler velvet.” (12) On the other hand, this was a small study, with only 12-13 men in each group. This means that further testing is needed to confirm the validity of the results.

Research has been conducted on the potential use of deer antler velvet for healing of wounds. In Russia (Arapov, 1969), Pantocrin was administered to patients with surgical and internal wounds. The study reported that there were a number of positive effects, including the normalization of arterial pressure, reduction in surgical complications, and becoming active quicker. In tests on rats (Wang, 1985), bone fracture repair was accelerated. A recent study by Bubenik found that antler helped heal epidermal wounds in rats. In another study by Takikawa, et al., researchers reported observing new bone formation following experimental whiplash injuries in rabbits. Pretreatment in rats reduced cell degradation and improved recovery times from extreme temperature and electric shock exposure.
New Zealand research reports that although the mechanism is unknown, deer antler velvet shows strong anti-inflammatory effects. Recent clinical tests suggest oral ingestion of glycosaminoglycan-peptide complex, or components such as chondroitin sulfate and glucosamine sulfate — both found in deer antler velvet — may help stimulate cartilage repair.
There is an increasing amount of scientific evidence supporting the benefits of deer antler velvet from decades of research carried out in Russia, Korea, China, Japan, Australia and New Zealand. An acute dose of 2,000mg/kg Deer Velvet Antler to rats (human equivalent dose of 320mg/kg) has failed to show toxic signs over 14 subsequent days of observation, and a 90 day trial with daily dosing of 1,000mg/kg did not show any significant toxicological symptoms of haemotological signs; a decrease in liver weight was noted in males, but under histological examination it appeared to be benign. Another rat toxicological study using 10% of the diet as Deer Velvet Antler during gestation and after birth noted that there were no apparent teratogenic effects on the rat pups and that serum AST (indicative of liver damage) was actually decreased 50% relative to control with no effect on γ-GT (another liver enzyme).
